I would like to stop the spindle motor at the end of a run with an M5. Does anyone have any suggestions on how to wire this up, and what type of relay can be used?
I am using Mach3, Xylotex board, & Rotozip router.

Can't help you with the relay, but you set up an output under ports and pins>Outputs, and then assign that output under ports and pins >Spindle setup.
This where a powered breakout board with direct interface to the Xylotex and buffered relay lines and high current relays is an advantage. Be aware that a lot of routers that use soft start will not work with an SSR (Solid State Relay). Good old electro-mechanical relays work every time!
