@cncman172 - I put the estops inline after the off button to kill the main power contactor that powers the 48v power supplies for the drives. Is this the right spot? I have seen some setups use an input on the MB3 board but I am not sure how that works.

The way you show your estop which kills source power to contact or will work. You diagram suggests your spindle is 110v is that correct. I have the same breakout board as you with the ESS

Yes, the "spindle" is just a Porter Cable 3.25 HP router at the moment. I have aspirations to upgrade to an ATC spindle one day but for now this will have to work. I have seen some wiring diagrams that use the estop with a pin on the MB3. How is yours wired?

Hi,
the only thing I see that is missing is a catch diode on the spindle relay. The inductive voltage spike when the relay de-energises will wreck the output of the MB3,
and really spoil your day.

In addition to the diodes on the relay to avoid spikes, I would also add EMI filters on the incoming AC power, to avoid EMI interference which will drive your machine crazy, with all kinds of unexpected motion.

@craig - I have no idea what a catch diode is, but Google did. So it acts as a one-way gate to not allow voltage back into the controller board from the relay? If you don't mind, can you explain where in the circuit this would be wired and what size I would need for a 15a 100v motor on a relay?

Yes, that filter will work fine

That filter will work but its only single stage, it should be two stage.

NO! Do not put a diode anywhere near the spindle.
A catch diode goes on the coil terminals of the relay.

Hi,
The catch diode goes between the coil terminals of the relay. Note the polarity, its important.

To recap:
a friend and I built 17 years ago. it sat at his place for 15 years unused. I recently went and picked it up and began the process of bringing the phoenix back from the ashes.
I completely replaced the control electronics including an ESS Smoothe Stepper with an MB3 Break Out Board.
The wiring is complete (I hope) and I have installed Mach4 and began configuring it. I have got all of the limit and E-stops configured but I cannot get the steppers to move.
A couple of notes:
I do not know anything about the stepper motors except they are NEMA34 and were purchased 17 years ago
I purchased new stepper drivers DM556T from Steppers online and have them installed and wired to the motors
The drives have holding power but will not move
The step and direction pins light appropriately when using the jog screen in Mach4
There is 48v power on the outside pins of the outputs for the stepper drivers (one for each winding)
Any help in diagnosing this would be appreciated.

Look at the manual for the drivers. Many stepper drivers have some pins to enable them. You usually need to ground the enable signal but check your manual
