Maybe the spindle is not connected to the board, in that case you cannot operate it from Mach3 but only from the front panel.

I confirm that the connector next to the lpt port is for Zero probe. You need only the two lower pins, pin 13.

Use the Mach3 post processor, for 3 axis works without need to modify, for 5 axis you need to modify it. Search the web, you will find it.
