I'd like some advice on changing a router over from NCStudio using a Weihong PCIMC card to a Mach3 system using either a breakout board & parallel port or a smooth stepper board.

The attached file shows what I have to work with and what I know of the existing setup.
Can I just swap existing 15 wires into a breakout board or a smooth stepper and then configure Mach3 to suit?

Any enlightenment greatly appreciated.

I would just keep the drivers and buy a BOB. I would suggest a PMDX-126 with the 107 spindle control board. This will be easy to hook up and work with Mach3.
